🚀 We'd like to spotlight this new excellent TCE review article. One of major side effects associated with T cell engagers (TCEs) is toxicities related to cytokine release syndrom. One key strategy to mitigating this risk is affinity attenuation of CD3 binding arm, in order to achieve potent tumor cell killing with minimal cytokine secretion. This review article provides an overview of the current state and recent developments in CD3 affinity-attenuation, both preclinically and clinically, with a focus on the challenges of developing TCEs with attenuated affinity to CD3 as well as identifying possible areas of improvement. Methods of Nanoparticle Synthesis Nanoparticles play a pivotal role in advancing medicine, catalysis, energy technologies, and environmental applications. Their physical and chemical properties are strongly influenced by the method of synthesis. This infographic outlines three primary approaches to nanoparticle synthesis: Chemical synthesis – relies on controlled chemical reactions and surface modification to produce stable nanoparticles with tailored properties. Physical methods – include processes such as laser ablation, where high-energy interactions generate nanoparticles of different shapes and sizes. Biological synthesis – an environmentally friendly route that utilizes plants, microbes, or enzymes for sustainable nanoparticle production. Each method provides unique advantages, enabling nanoparticles with specific morphologies and functionalities. Proud to share our recent success in scaling up the production of a new #adjuvant — a crucial step toward enabling the development of next-generation vaccine adjuvants. At the IAVI Product Development Center (PDC), we bridge the gap between innovation and access by advancing manufacturing solutions for vaccines, adjuvants, and monoclonal antibodies targeting diseases that disproportionately impact LMICs. In this collaborative effort, IAVI’s Sam Pallerla (Sr. Director, Product Development and Manufacturing) led a cross-functional product development team, including partners and stakeholders from Massachusetts Institute of Technology, Scripps Research, and The National Institutes of Health, working closely with several CDMO partners. Together, we created and demonstrated a GMP-compliant, scalable process for producing a novel adjuvant that combines saponins and monophosphoryl lipid A (MPLA). This formulation has strong potential for use across various vaccine platforms and supports the field’s progress toward globally accessible, transparent, and cost-effective adjuvant technologies.
