Scopo del videogioco è battere un'avversaria (scelta fra otto possibilità) a poker. Ogni puntata vengono puntati dei capi d'abbigliamento ed alla fine, se il giocatore vincerà la partita, l'avversaria si ritroverà nuda. Il giocatore può scegliere fra otto diverse avversarie: Maria Whittaker, Amanda Godden, Ginny Champ, Donna Ewin, Trine Michelsen, Sofia Bratlund, Jane e Signe Andersen. A differenza di molti giochi simili Cover Girl Strip Poker mostra delle sequenze animate fra una immagine e l'altra delle fasi di spogliarello delle avversarie del giocatore, benché presenti in solo quattro delle otto protagoniste femminili.
