Collective unconscious: How gut microbes shape human behavior

The human gut harbors a dynamic and complex microbial ecosystem, consisting of approximately 1 kg of bacteria in the average adult, approximately the weight of the human brain. The evolutionary formation of a complex gut microbiota in mammals has played an important role in enabling brain development and perhaps sophisticated social interaction. Genes within the human gut microbiota, termed the microbiome, significantly outnumber human genes in the body, and are capable of producing a myriad of neuroactive compounds. Gut microbes are part of the unconscious system regulating behavior. Recent investigations indicate that these microbes majorly impact on cognitive function and fundamental behavior patterns, such as social interaction and stress management. In the absence of microbes, underlying neurochemistry is profoundly altered. Studies of gut microbes may play an important role in advancing understanding of disorders of cognitive functioning and social interaction, such as autism.

This excellent new paper by Kronsbein et. al. describes studies on advancing forced degradation studies: design of experiments for enhanced structure-function relationship analysis in biotherapeutics. Quoting from the abstract: "Understanding the relationship between structure and function is crucial in drug development. Recombinant proteins present unique challenges when establishing structure–function relationships (SFR) due to the extensive options for potential modifications across numerous residues. Even simple stress studies lack degradation selectivity as they lead to multiple, simultaneous modifications. Co-occurring modifications at amino acid residues make direct correlations with a specific stressor and resolving the causal impact on functionality difficult. The introduction of more variance into the data set would reduce the correlation structure. In this study, we demonstrate the feasibility and benefits of a multifactorial design approach for SFR establishment. Design of experiments (DoE) allows for the parallel investigation of stress factors via combined experiments, resulting in a higher variance of stress conditions and consequently a broader variation in degradation. This approach enables a more insightful correlation analysis along with model-based data evaluation strategies, thereby facilitating significantly improved data interpretation results during SFR studies."

In Nature today my lab led by Djuna von Maydell dissects how ABCA7 loss of function variants contribute strongly to Alzheimer’s disease risk, including disruption of mitochondrial function and lipid metabolism in excitatory neurons from human brain samples. Importantly, after observing disrupted phosphatidylcholine metabolism in iPSC cultures engineered with ABCA7 variants, we treated cells with CDP-choline and found this rescued the metabolic deficit, reversing many of the disease-associated abnormalities associated with the variants, including amyloid secretion and neuronal hyper excitability. The study strengthens the evidence base suggesting that disruption of lipid metabolism plays a key role in the pathophysiology of Alzheimer’s disease. Restoration of Lysosomal Hydrolase Activities by LRRK2 Inhibition in GBA1- and LRRK2-Associated Parkinson's Disease Patient-Derived Cells A recent study published in the Journal of Neurochemistry https://lnkd.in/eCQSps5W explored the therapeutic potential of LRRK2 inhibitors for both GBA1 and LRRK2-associated Parkinson's disease (PD). Researchers evaluated the effects of LRRK2 inhibition on lysosomal hydrolase activity and alpha-synuclein levels in various patient-derived cell types, including PBMC-derived macrophages and iPSC-derived dopaminergic neurons. They discovered that LRRK2 inhibition enhances the activity of multiple lysosomal hydrolases, including glucocerebrosidase (GCase), and promotes GCase's translocation to lysosomes. This effect was notably more pronounced in cells from GBA1-PD patients than in those from LRRK2-PD patients. Additionally, the study found that LRRK2-PD patient macrophages exhibited reduced galactosylceramidase (GALC) activity and elevated levels of lysosphingolipids, which are known to stabilize neurotoxic alpha-synuclein.
