  • Inherited erythromelalgia due to mutations in SCN9A: natural history, clinical phenotype and somatosensory profile

    Brain

Inherited erythromelalgia, the first human pain syndrome linked to voltage-gated sodium channels, is widely regarded as a genetic model of human pain. Because inherited erythromelalgia was linked to gain-of-function changes of sodium channel Na v 1.7 only a decade ago, the literature has mainly consisted of reports of genetic and/or clinical characterization of individual patients. Because inherited erythromelalgia was linked to gain-of-function changes of sodium channel Na v 1.7 only a decade ago, the literature has mainly consisted of reports of genetic and/or clinical characterization of individual patients. This paper describes the pattern of pain, natural history, somatosensory profile, psychosocial status and olfactory testing of 13 subjects with primary inherited erythromelalgia with mutations of SCN9A , the gene encoding Na v 1.7. Subjects were clinically profiled using questionnaires, quantitative sensory testing and olfaction testing during the in-clinic phase of the study.

  • Pattern of response in patients with moderate-to-severe psoriasis treated with etanercept.

    British Journal of Dermatology

Etanercept (ETN) 50 mg once-weekly (QW) or 50 mg twice-weekly (BIW) for 12 weeks, followed by 50 mg QW in all subjects through week 24 improved psoriasis in patients with concomitant psoriatic arthritis in the PRESTA trial. Data from PRESTA were used to evaluate the effect of ETN in the treatment of psoriasis by Psoriasis Area Severity Index (PASI) body-region and component, and determine if PASI responses correlate with the Dermatology Life Quality Index (DLQI).

  • Practical Application of the Prentice-Wilcoxon Test for Paired Time-to-Event Data

    PSI Conference Poster

In survival analyses we conventionally compare (two or more) strata with regards a time-to-event endpoint; patients are either represented in one strata or the other and the strata are independent of each other.
In this poster we analysed data where we are interested in the within-patient time-to-improvement of psoriasis symptoms in various parts of the body; within-patient data are not independent, and conventional survival analysis methodology is therefore not appropriate. A…

In this poster we analysed data where we are interested in the within-patient time-to-improvement of psoriasis symptoms in various parts of the body; within-patient data are not independent, and conventional survival analysis methodology is therefore not appropriate. A Prentice-Wilcoxon Test allows paired/tied/correlated data within a patient to be compared.
The endpoint analysed was PASI75 – the time to 75% improvement in PASI. PASI is a composite endpoint capturing the extent and severity of symptoms in psoriasis. This poster examined the practical application of the Prentice-Wilcoxon test, its ease of use and discussion of its merits.

  • Complex Randomisations Need Not Be Complex

    PSI Conference Poster

In collaboration with one of our clients, we designed a trial with five treatment arms: three were various doses of the active compound, one arm was a positive control and the final arm was a negative control. It was desired to have an unequal allocation to treatment: 3:3:3:2:2, enrolled at two sites; patients were additionally stratified according to a baseline (binary) characteristic. An interim analysis was planned, and we needed to ensure that at any snapshot in time, the allocation ratio…

An interim analysis was planned, and we needed to ensure that at any snapshot in time, the allocation ratio adhered to the 3:3:3:2:2 as closely as possible. This poster examined why we would have unequal allocation to treatment in the first place, and how to go about performing a randomisation such that for any interim snapshot, we could ensure as balanced as possible allocation overall. Randomisation within sub-blocks is the solution, where the sub-blocks are randomly ordered within the main block.

  • Comparative growth performance of pigs immunised with a gonadotrophin releasing factor vaccine with surgically castrated pigs and entire boars raised under conventionally managed conditions.

    Berl Munch Tierarztl Wochenschr.

The objective of this study was to determine the effect of using a gonadotrophin releasing factor (GnRF) vaccine on growth performance of male pigs. Vaccinated pigs were compared with surgically castrated pigs and entire males under conventionally managed farm conditions.

  • Effect of a gonadotropin-releasing factor vaccine on follicle-stimulating hormone and luteinizing hormone concentrations and on the development of testicles and the expression of boar taint in male pigs.

    Theriogenology

The objective of this study was to determine the effect of using a gonadotropin-releasing factor (GnRF) vaccine on follicle-stimulating hormone (FSH) and luteinizing hormone (LH) concentrations in plasma, the size of testicles, and the expression of boar taint in male pigs. Vaccinated pigs were compared with surgically castrated pigs and entire males.
