For 35 years PBL Assay Science has developed and supplied specialty immunoassay products and biomarker testing services to basic, drug development, translational, and clinical researchers around the world. Interferons and other cytokines are immune system proteins and important biomarkers in drug discovery and development. PBL has built a reputation as a high-quality manufacturer of interferon and cytokine proteins and antibodies, as well as pre-packaged, best-in-class interferon and cytokine immunoassay kits. 𝗪𝗵𝘆 𝗱𝗼 𝗱𝗿𝘂𝗴𝘀 𝗯𝗲𝗵𝗮𝘃𝗲 𝗱𝗶𝗳𝗳𝗲𝗿𝗲𝗻𝘁𝗹𝘆 𝗶𝗻 𝗹𝗮𝗯 𝗺𝗼𝗱𝗲𝗹𝘀 𝗮𝗻𝗱 𝗶𝗻 𝗽𝗲𝗼𝗽𝗹𝗲? A new study in PLOS Computational Biology explored how interferon-alpha (IFN-α) behaves across three settings — in vitro, in vivo (mice), and in silico (QSP models) — revealing key translational gaps in how we interpret immune signaling data. 🔹 𝗜𝗻 𝘃𝗶𝘁𝗿𝗼 = 𝗶𝗻 𝘃𝗶𝘃𝗼 Cell culture studies often overestimate drug potency. This work showed that IFN-α responses in vitro can be up to 16× stronger than what’s observed in living mice — largely because in vitro systems lack clearance and feedback regulation. 🔹 𝗠𝗼𝘂𝘀𝗲 = 𝗵𝘂𝗺𝗮𝗻 Even with similar doses, murine antiviral responses were predicted to be 8–16× stronger than in humans due to more efficient receptor binding. The study underscores why mouse data must be carefully contextualized before extrapolating to patient outcomes. 🔹 𝗤𝗦𝗣 𝗺𝗼𝗱𝗲𝗹𝗶𝗻𝗴 𝗯𝗿𝗶𝗱𝗴𝗲𝘀 𝘁𝗵𝗲 𝗴𝗮𝗽 By integrating pharmacokinetics (PK) and pharmacodynamics (PD) into a unified quantitative systems pharmacology (QSP) model, researchers created a powerful translational framework. Such models can guide dosing decisions, refine preclinical study design, and improve cross-species predictions during drug development. To support these experiments, murine IFN-α4 from PBL Assay Science was used as part of the study’s validation work.
