6. Manufacturing Procedure
ProRenew Complex CLR is obtained by means of fermentation of Lactococcus lactis. After
growth is complete, the bacteria are disintegrated by means of high-pressure
homogenization, thus releasing the cytoplasm fractions and cell wall components. No
fractions are isolated after cell disintegration, thus ensuring the presence of all components in
ProRenew Complex CLR in their natural distribution.

Recommended Use:
Recommended for products that aim for stimulation of skin's renewal processes. Both
accelerating it and improving its quality. ProRenew Complex CLR can be used in formulation
concepts to accelerate the recovery of skins barrier function, regardless of type of impairment
(environmental stress, aging).

Preferably used in emulsions but also in gel-type type formulations or serums, which are
stabilised with suitable hydrocolloids or polymers.

Acute Toxicity
ProRenew Complex CLR is based on ingredients, which have a long tradition in food industries
as well as in cosmetic applications. It is considered to be non-toxic and safe in cosmetic
products usage at the levels advised by us.

Primary Skin Irritation


A 48 hour Patch test with a 50% solution of ProRenew Complex CLR has been performed on a
panel of 50 volunteers (18-67 years; 24 normal healthy, 3 eczema, 3 allergy and 20 subjects
with sensitive skin). All participants completed the study. Under the test conditions, SDS (1% in
water) caused positive reactions in 14 subjects. The negative control water showed no

reactions. None of the subjects showed any reaction to the test product. On the basis of these
test results and under the test conditions, the product "ProRenew Complex CLR " is to be
classified as 'harmless' with regard to the possibility of skin irritation.

Eye irritation potential


ProRenew Complex CLR was tested undiluted using the Bovine Corneal Opacity and
Permeability Assay (BCOP) assay. The calculated mean in vitro irritation score was: 0.97.
According to the evaluation criteria ProRenew Complex CLR is classified into UN GHS “No
Category”. Therefore, ProRenew Complex is not posing a risk when applied in formulations
to the eye area region.
ProRenew Complex CLR was additionally tested, undiluted, on the chorioallantoic membrane of
the hen's egg where it produced an irritation index of 0.00. It therefore was classified as "non-
irritant".

Experienced Skin Tolerance under Use Conditions


Skin incompatibilities have not been reported.

Sensitization
Human Repeated Insult Patch Test (HRIPT):
ProRenew Complex CLR was tested on 50 volunteers (repeated application) using a 50%
solution of the active in 0.9% NaCl buffer. The methodology used corresponds to the guideline
« Test Clinique Final de Securité d’un Produit Cosmétique en vue de confirmer son absence de
potentiel sensibilisant cutané retardé: Recommandations aux promoteurs de recherche et aux
prestataires de service », from AFSSAPS. Two study phases, induction phase followed by
challenge phase, allow to conclude on the skin compatibility and cumulative irritation of the
product and its allergenic potential.
The repeated application of 50% ProRenew Complex CLR solutions allowed to conclude that
this product has a very good skin compatibility and has no allergenic potential (supporting
hypoallergenic claims).

the bacterial reverse mutation assay using 4 different strains of Salmonella typhimurium and
E. coli WP2 uvrA bacteria, with and without metabolic activation. No mutagenic effects could
be observed in any of the five tester strains up to the highest dosage.

Mode of action
ProRenew Complex CLR is produced by fermentation of probiotic bacteria in a bioreactor, with
subsequent inactivation and mechanical disintegration.

Probiotic bacteria are well-established in the food industry and their benefits for the human
body are described in many scientific, peer-reviewed papers. In the gut they beneficially
influence the composition and metabolic activity of the endogenous bacteria and some
probiotic strains are even able to inhibiPt the growth of pathogenic bacteria. Probiotic bacteria
are also reported to be able to modulate the immune system, essentially improving
immunocompetence and, therefore, our body's ability to adapt to negative influences.
Interestingly, it is not the whole living probiotic bacterial cell which is needed for the latter
activity, it is the constituents and metabolites from these bacteria which are essential. A lysate
of Lactococcus lactis, as in ProRenew Complex CLR, essentially containing the cell debris of this
bacterium, has shown in the studies performed by CLR GmbH, that it is able to improve both
the skin renewal processes and the processes involved in the skin's adaptation to negative
influences.

Processing
The microorganism fragments contained will form a sediment after some time of storage.
Shake or agitate before use to obtain homogenous solutions. When formulating ProRenew
Complex CLR it has to be considered, that electrolyte sensitive components of formulations
might be influenced. Electrolyte sensitive rheology modifiers and hydrocolloids might lose some
efficacy, depending on the percentage of ProRenew Complex CLR used in final formulations.

During the production process ProRenew Complex CLR should not be exposed to temperatures
exceeding 40 °C. The pH of final formulations containing ProRenew Complex CLR should range
between 4 and 7.

Storage
At room temperature (15 - 25 °C), in well-closed containers, protected from light.
ProRenew Complex CLR should not be stored for longer than 1 year before processing. Once
opened, the packs should be fully used. Any remaining product should be discarded.
