AEC
Introduction:
Definition: An absolute eosinophil count is a
blood test that measures the number of one type of
white blood cells called eosinophils.
 Eosinophils become active when you have certain
allergic diseases, infections, and other medical
conditions.
AEC
 Aim: To determination of absolute Eosinophil count
 Clinical Significance: Eosinophil count has a close
relationship with the adrenal function & Eosinophil count can
be diagnostic for adrenal related disorder. Increased
Eosinophil count is often associated with allergic condition,
parasitic infection & certain leukaemia etc. Increase in the
adrenal function (hyperadrenalism or Cushing's syndromes)
is associated with a fall in Eosinophil count.
 Method: Haemocytometry method
AEC
 Normal Range: 40 - 440 /cumin
 Principle: AEC diluting fluid is called Hingleman's
solution. Blood is diluted 1:10 with a special diluting
fluid, which remove red cells & stains the eosinophil red.
These cells are then counted under low power in a
known volume of fluid by using counting chamber.
 Requirements: Microscope, Neubauer's chamber, Hb
pipette, cotton Test tube, diluting fluid Diluting fluid
contains: - Yellow eosin (0.5g), 95% phenol (0.5m1),
formalin (0.5m1), Distilled Water (99m1).
 Sample: EDTA blood sample
AEC
 Procedure:
1. Identify the right patient.
2.Collect all the requirements.
3. Do the labelling.
4. Collect require amount of blood by capillary or vein puncture.
5.In a labelled test tube take 0.36 ml of AEC diluting fluid.
6. Add 0.04 ml of blood (use Hb pipette, twice).
7. Mix it well & keep it for 10 min.
8. Charge the Neubauer's chamber & let it stand
under a moist Petridis for about 2 to 3 min.
9. Count the Eosinophil under the low power of microscope in all the nine
squares.
10. Do calculation.
11.Clean the working area
12. Put all the requirements at them of place
13. Wash the hand.
AEC
 Calculation:
Precaution: Eosinophil disintegrates in
the diluting fluid hence the count should
not be delayed for more than 30 minute
after diluting the blood.
