This document describes procedures for estimating the purity of magnesium sulfate and calcium gluconate. It first details the preparation of a 0.05 M disodium edetate solution and its standardization. For magnesium sulfate estimation, 0.3 g of the compound is dissolved and titrated against the disodium edetate solution. The volume used is used to calculate purity percentage. For calcium gluconate estimation, an accurately measured volume equivalent to 0.5 g of the compound is titrated against disodium edetate after the addition of magnesium sulfate and ammonia solutions. The volume used is then used to calculate the amount of calcium gluconate present.

Preparation of 0.05M Disodium Edetate
Dissolve 37.2g of disodium edetate in
sufficient water to produce 1000ml.
Standardization of 0.05 M Disodium
edetate
Pipette out 10 ml of 0.05 M CaCl2 solution
into a conical flask & 5 ml of Ph 10 buffer
solution. Add 2- 3 drops of solochrome
black T indicator& titrate with EDTA
solution. Repeat titration for concordant
value.

1) Estimation ofMagnesium
Sulphate
Weigh accurately 0.3 g of anhydrous magnesium
sulphate , dissolve in 50 ml water. Add 10 ml of ph 10
ammonia buffer & 50 mg mixture of mordant black II
& NaCl as indicator. Titrate with 0.05 M disodium
edetate until the solution becomes blue.
Each ml of 0.05 M Disodium edetate until the solution
becomes blue.
Each ml of 0.05 M Disodium edetate = 0.012325g of
MgSO4.
Percentage purity of MgSO4=
Volume of EDTA x factor x 100 x Molarity of
EDTA(actual)
Weight of MgSO4 in gm x molarity of EDTA (expected)

2) ESTIMATION OFCALCIUM GLUCONATE INJECTION
Calcium gluconate injection contains a
quantity of calcium equivalent to not less
than 8.5% & not more than 9.4% of stated
amount of calcium gluconate. Assay of
calcium gluconate is performed by
substitution reaction/replacement titration.

Procedure
To an accuratelymeasured volume
equivalent to 0.5 g of calcium gluconate,
add 50 ml of water & add 5 ml of 0.05 M
MgSO4 & 10 ml of strong ammonia
solution & titrate with 0.05 M disodium
edetate using mordant black II mixture
as indicator.
From the volume of 0.05 M disodium
edetate required subtract the volume of
the MgSO4 solution added.
